Locus Sequencing Kits provide reagents for amplification, purification, and Sanger sequencing of HLA alleles (variants) from DNA. Sequencing fragments produced using this CE Marked kit are subjected to capillary electrophore.
The AllSet+ Gold DNA Wipe Test is used to monitor DNA contamination that may interfere with the accurate reporting of AllSet+ Gold SSP and SeCore SBT test results. The test is designed to detect a majority of previously amplified, and non-amplified, human genomic DNA containing sequences specific to exon 2 and/or exon 3 of the following HLA genes: HLA-A, -B, -C, -DRB, -DQA, -DQB, -DPA, -DPB genes.
Made to order SeCore GSSP’s, the kits are intended for the identification and definition of Class I and II Human Leukocyte Antigens (HLA) for research applications. The SeCore HLA Sequencing and GSSP kits provides human histocompatibility information of HLA Class I (A, B and C) and Class II (DPB1, DQB1 and DRB) Loci using genomic DNA isolated from whole blood specimens.
Using the Sanger sequencing method, SeCore SBT sequencing kits provide bi-directional sequencing for Class I loci: HLA-A (exons 1 – 5), HLA-B (exons 1a – 5), and HLA-C (exons 1 – 7), and bi-directional sequencing for Class II loci: HLA-DRB1b (exons 2 – 3), HLA-DQB1 (exons 2 – 3), and HLA-DPB1c (exons 2 – 4). The uTYPE™ HLA Sequence Analysis Software is used to evaluate the results against the IMGT/HLA database and to help determine the molecular typing.

High throughput HLA A Locus typing kit via sequencing based technology for labs handling batches of approximately 300 samples of a single locus.
SeCore Sequencing Kits directly identify DNA sequence from target HLA genes. Locus specific DNA amplification is accomplished by cycling an amplification mix, FastStart™ Taq DNA Polymerase and sample genomic DNA. The resulting product is treated with ExoSAP-IT™ prior to sequencing, to degrade the unincorporated primers and free nucleotides. The nucleotide chronology and resulting HLA subtype is determined by multicolor, fluorescence-based sequencing.
